Culture of Bovine Retinal Pigment Epithelium : Topographical Differences of Morphological and Growth Rate in Vitro


Abstract
To evaluate pathogenesis of age-related macular degeneration, cell cultures of adult bovine retinal pigment epithelium(RPE) were propagated from central and peripheral regions of the same eyes to study topographical differences in cell growth and
to
compare the differences of growth rate between two areas.
@ES The results obtained were as follows :
@EN 1. A regional variation in the morpholgy of bovine retinal pigment epithelium of central and peripheral regions was observed. The retinal pigment epithelium frcm peripheral region appeared brown to black color due to high melanin granule
content but
retinal pigment epithelium from central region appeared more whitish due to low melanin content. Attachment of retinal pigment epithelium from central region to culture dish was slower(average 4 days) than those of peripheral region(average 3.5
days).
2. The growth rate of retinal pigment epithelium from central and peripheral region during generation was observed.
It declined with serial passage in culture. The growth rate of retinal pigment epithelium from peripheral region at first generation was the highest. And there was a statistical differences in growth rate with passing in generation(P<0.05).
